In fact, the specific reason is also very simple, it may be that the quality of the coloring solution is not good enough. In general, the decline in the OD value after termination of the reaction is not very obvious. After termination, the absorbance value will decay with time, so it is generally measured immediately after the addition of the stop solution. 1 Color time adjustment, if your current color development time is 10MIN, then adjust to 30MIN, add stop liquid, observe whether the above phenomenon appears.
2 Add a small amount of glycerin to the stop solution. It is recommended that you use the RD brand ELISA kit, the color development time is 30 minutes, and it is required to test within 30 minutes after termination. After adding the stop solution, it is best to test it within 2 to 3 minutes after adding the stop solution. This is the OD value. Relatively high. The fitted value can reach four 9.
