China has developed a new technology for nano-enzyme treatment of tumors

Recently, Nature Communications magazine published the latest research results of nano-enzyme catalytic treatment of tumors by the Gao Lizeng research group of Yangzhou University Medical College and the research group of the Institute of Biophysics of the Chinese Academy of Sciences. This study demonstrates for the first time that by regulating the in vivo activity of the nanoenzyme multi-enzyme system, tumor metabolites can be catalyzed as toxic substances to achieve specific killing of tumors. According to reports, the study is the first to use nanozymes directly in the treatment of tumors.

The research team designed and invented a novel carbon-nitrogen nanozyme with multiple enzyme activities, and utilized ferritin navigation to achieve precise delivery and enzyme regulation of nanozymes in vivo, using tumor-specific metabolites to play tumor-specific. Efficient killing effect, established a new strategy of nano-enzyme targeted catalytic treatment of tumors, providing new ideas and techniques for cancer treatment.

According to reports, tumor metabolism has different characteristics from normal cells. How to use tumor metabolites to develop "the way to the other, to make the body" is a new field of tumor biotherapy. Since hydrogen peroxide can be catalyzed by peroxidase to produce cytotoxic free radicals, peroxidase is expected to develop new drugs for tumors. However, natural enzymes tend to be less stable and are easily deactivated in complex in vivo environments, and their application is limited. Researchers in this work have taken a different approach and explored the application of nanozymes with both enzyme catalytic activity and nanomaterial stability in this strategy.

The researchers first designed and synthesized a novel nanozyme with four enzyme activities: oxidase, peroxidase, catalase and superoxide dismutase. This nanoenzyme multienzyme activity is achieved by doping the mesoporous carbon sphere with nitrogen. How to accurately deliver the nanozyme to tumor cells and specifically activate its oxidase and peroxidase activities to produce reactive oxygen species is the key to determining whether carbon-nitrogen nanospheres can be used in tumor therapy.

The researchers used ferritin to modify the novel carbon-nitrogen nanozyme, and experimentally verified that the ferritin-modified carbon-nitrogen nanozyme can specifically recognize tumor cells and locate them in the lysosomes with acidic environment inside the tumor cells. Its oxidase and peroxidase are specifically activated, catalyzing the oxygen and hydrogen peroxide in the tumor to produce highly toxic reactive oxygen free radicals, achieving specific killing of tumor cells. Animal tumor-bearing experiments showed that ferritin-carbon nitrogen nanozyme can significantly inhibit tumor growth.
