Application of Circulating Ultrasonic Extraction Technology in Deep Processing of Chinese Medicinal Materials

The circulating ultrasonic extraction technology and device have been successfully used for the extraction of natural products from dozens of Chinese herbal medicine raw materials. Some of the results are as follows:

(1) Terpenoids, lactones

Artemisinin is generally extracted by petroleum ether, gasoline cold dipping or stirring at 50 ° C or lower, the extraction rate is about 60%, and the extraction time is 24-48 hours. The artemisinin was extracted with HF-2B, and the effects of fluid circulation flow, ultrasonic time, ultrasonic intensity, solid/liquid ratio, extraction temperature and agitation speed on the extraction rate of Artemisia annua L. and petroleum ether loss were studied. Under the experimental conditions, the extraction rate of artemisinin was over 90%, the recovery rate of artemisinin was increased by more than 25% compared with the conventional extraction method, the extraction time was shortened to 20 minutes, and the recovery rate of petroleum ether was 90%, which was significantly improved compared with the conventional extraction method. .

(2) Flavonoids

In the extraction of flavonoids from Ginkgo biloba, the extraction rate of conventional boiled for 4 hours was 56.5%; at room temperature, extraction with HF-2B for 10 min, the extraction rate was 82.6%, extraction was 30 min, the extraction rate was 88.3%; at 40 ° C, HF-50G was used. The extract was extracted by a circulating ultrasonic extractor for 30 min, and the extraction rate was 91.6%, which was higher than that of the conventional ginkgo flavonoid extracting rate at 100 °C for 6 hours.

When the propolis flavonoids were extracted, the extraction was carried out for 48 hours, and the extraction rate was 36%. The extraction rate was 41.8% after extraction with HF-2B for 30 minutes at room temperature.

At 30 °C, 50% ethanol HF-2B was used to extract Epimedium for 30 minutes, and the flavonoid extraction rate was 87%.

When flavonoids were extracted, HF-20B was used for 60 min. The total flavonoid yield was 11.5% in 80% ethanol and 8.8% in 85% ethanol.

(3) Alkaloids

When the betaine was extracted from Cistanche deserticola with water, extraction with HF-2B for 20 minutes was equivalent to the extraction of betaine by conventional leaching for 24 hours and 100 °C reflux for 6 hours.

Using HF-2B as the extractant, extract the total alkaloids from the sand oysters, extract them by ultrasonic extraction at room temperature for 30 minutes, extract twice, and the extraction rate is more than 85%. The three effects are better; the extraction is 4-6 than normal temperature. Hour/time, the effect of extracting three times is better, and the effect is higher than that of diafiltration for 24 hours; with chloroform as extractant, ultrasonic extraction is 7 times, which is better than conventional continuous extraction for one week.

(4) mites

When Cistanche tubulosa was extracted from Cistanche deserticola with methanol, extraction with HF-2B for 30 minutes was equivalent to the extraction rate of Cistanche deserticola with conventional leaching for 24 hours and reflux at 80 °C for 5 hours.

At 30 °C, extracting Epimedium with 50% ethanol HF-2B for 30 minutes, and the rate of cumin foxtail was 90%.

(5) Polysaccharides

When the polysaccharide was extracted from Cistanche deserticola, the polysaccharide extraction was carried out using HF-2B for 20 minutes, which was higher than the conventional extraction for 24 hours and 100 ° C reflux for 6 hours.

When sulphate polysaccharide was extracted from kelp by HF-2B, it was extracted at 30 ° C for 20 minutes to obtain the same sulfate ester polysaccharide extraction rate by boiling method (100 ° C) for 5 hours.

(6) Tea polyphenols

Tea polyphenols and catechol were extracted from tea leaves, and the total amount of tea polyphenols and catechins was increased by 40% compared with 100 °C water extraction for 30 min at room temperature. The obtained polyphenols and catechol samples had no traits. The difference does not change the structure of the components of tea polyphenols and catechol.

(7) Solvent extraction and extraction and purification in slurry extraction

The HF-2B was used to extract the sulfated polysaccharides from the solvent. It is hoped to realize the coupling and integration of low-micro content natural active ingredients extraction, filtration, concentration, crude extraction, separation and purification, and directly obtain the purified products. Dip. At room temperature, the ultrasonic action time is 30 minutes, the extractant concentration is 7%, etc., the polysaccharide extraction rate can reach above 92%, the fucose content in the polysaccharide is ≥35%, and the pharmaceutical polysaccharide is obtained in one step (fucose content ≥30) %) requires that the cost is only about a quarter of the regular extraction.

(8) Other

The pepper was boiled with 75% ethanol for 8 hours and the absorbance was 2.274; the temperature was extracted with HF-2B for 30 min at room temperature, the absorbance was 1.993, the extraction was 40 min, and the absorbance was 2.053.

The above test results show that the circulating ultrasonic extraction device has good amplification performance, and the same equipment selection may have a large difference in results.
