In the Ningxia region, naturally bred fry larvae typically become available in mid to late May, resulting in smaller fish sizes and lower survival rates during winter. To address this issue, local squid farmers have traditionally used air-fry from southern Wuzi, but this method is expensive and increases the risk of pathogen introduction. In 2004, an artificial breeding experiment was conducted in March and April to enable earlier fry release, aiming to boost production and income. The experiment began with broodstock selection and cultivation. The breeding ponds were 8 acres in size, covered with rectangular plastic greenhouses, with a sludge depth of 10–15 cm and water depth of 1.2 m. Before stocking, the ponds were treated with dry lime (100–150 kg/mu). Broodstock, consisting of sturgeon aged 2–8 years and weighing 1–4 kg, were stocked at a density of 50–80 tails per mu. They were fed concentrated feed 1–2 times daily, at 5–10% of their body weight. After 30 days of intensive feeding, gonads developed well. Broodstock selection focused on individuals with no injuries, smooth bodies, and active behavior. Females had large, soft bellies with visible ovaries, while males had smaller abdomens. Artificial oxytocin (DOM+LRH-A2) was injected into females at a dose of 3 mg DOM + 10 μg LRH-A2 per kg, with males receiving half the dose. Injection was performed carefully to avoid internal damage. Artificial insemination was done using dry methods. Eggs were collected by gently squeezing females and male sperm was added to fertilize them. Fertilized eggs were then placed in muddy water to adhere before being transferred to hatching tanks with running water. The hatching rate was around 80%, though it dropped slightly in later batches due to residual enzymes in the eggs. Pre-cultivation involved preparing ponds with lime and fermented manure to enrich the water. Fry were stocked at 120,000–150,000 per mu, and fed egg yolk and soy milk. Larvae were monitored regularly for disease and growth. The survival rate of fry reached 95.7%, with over 3 million fingerlings reaching 24 mm or more. The study concluded that using plastic greenhouses for broodstock rearing and controlled temperature incubation allowed early propagation of pufferfish, improving economic returns. Proper timing, water temperature control, and disease prevention were key to success. Future efforts should focus on optimizing broodstock age and feeding to enhance fertility and production.
